
Ole J. Bjerrum
(MD, DMSc), served at the Medical Faculty, University of Copenhagen from 1969-1987. Further to this Ole J. Bjerrum has 14 years of industrial experience at Novo Nordisk, working as director first for a diagnostic unit then with various aspects of drug discovery and finally as liaison officer between the company and academia and national and international research organisations. In 2001, Professor in Pharmacology at the Danish University of Pharmaceutical Sciences. Now department of pharmacology and pharmacotherapy, Faculty of Health and Medical Sciences, University of Copenhagen. Scientifically Ole J. Bjerrum has been engaged in the analysis and characterisation of membrane receptors and transporters (thesis 1977). Lately, he has engaged himself in in vitro and in vivo pharmacology aspects of pain with a translational perspective utilizing the combination aspect of registrered drugs. Through his membership of EUFEPS, where he now is past-president, he has engaged in three workshops related to the problems of European drug development covered under the title New Safe Medicines Faster, which paved the way for the EU programme Innovative Medicines Initiative worth 2 billions Euro.
Research Area:
Translational pharmacology/medicine with focus on neuropathic pain. Patophysiological investigations through pharmacological interventions.
Background and aim:
Pain after nerve injury (neuropathic pain) appears infrequently among patients. Further the condition is difficult to treat. Preclinical and clinical experiments with analgesics have shown individual discrepancy in respons. This probably reflects patient variation in the activation of the pain provoking mechanisms of sensing, transduction and transmission. The aim of the project is through better knowledge of the patophysiological mechanisms to improve the individual treatment regime of patients suffering from chronic pain.
Methods
Spared Nerve Injury (SNI) models (SNI, SNL, CCI) in rats for neuropathic pain. Rat models for acute and inflammatory pain (Hot plate, test with formalin and complete Freund's adjuvant, Hargreaves plantar test. Pharmacodynamic pain assessment with von Frey filaments, pin prick Randa Solito.
Cell culturing of neurons from dorsal root ganglia (DRG). Fluorescent imaging af Ca++ fluxes in cultured DRG neurons in presence of pharmacological active compounds. Immunostaining of cell markers of cultured neurons. Pharmacological characterisation of individual neurons in DRG cell cultures from naïve/injured/treated SNI, SNL rats.

New treatment regimens for neuropathic pain tested on rat pain models.
§ Effect of combination therapy of drugs acting on different targets of the pain transmission pathway inclusive PK/PD studies.
§ Effect of pharmacologic prophylaxis for the development of neuropathic pain.
